Abstract

The fidelity of DNA Polymerase I, isolated from B. licheniformis and B. stearothermophilus, was tested in the presence of carcinogens and anticancer compounds. All of these compounds inhibited (62.4-99.2%) the synthesis of DNA using Poly(dA-dT) as a template. The compounds were inhibitory at all concentration levels tested, 0.01, 1.0 and 100 mM. The fidelity of the enzyme appeared to be enhanced by the presence of these compounds, as indicated by misincorporation (50-70%), that is, the incorporation of incorrect nucleotide. Misincorporation in the presence of either carcinogens or anticancer compounds was less than that of control. The results from both organisms showed similar trends.
