Abstract
The analysis of proteins by Desorption Electrospray Ionization (DESI) is challenged by a mass depend loss in signal response. We show that this loss in sensitivity originates primarily from the incomplete dissolution of proteins into desorptiopn spray solvent. The addition of ammonium bicarbonate is used to improve protein dissolution and we demonstrate beneficial effects this has on protein detection by DESI-MS. We demonstrate improved signal to noise ratio's for all proteins investigated, up to 66kDa. In addition to improvements in signal to noise ratio, changes in charge states were also observed and are described.
